We used mouse dermal fibroblasts 13 deficient in Ku70 and p53, either complemented by WB analysis using 15421-1-AP. Various lysates were subjected to SDS PAGE followed by western blot with 15421-1-AP (ATP citrate lyase antibody) at dilution of 1:2000 incubated at room temperature for 1.5 hours. Excision of AP sites near DSBs by a 5'dRP/AP lyase is thus critical for efficient and accurate resolution of such ends by cellular NHEJ (Figure 1a). Consistent with other studies 14 , 15 (reviewed in 16 ) an alternative to NHEJ (Alt-NHEJ) allows for significant joining of undamaged ends in Ku70 deficient cells ( Figure 1d , columns 1 and 2, 7 and 8).

In Arabidopsis, AtOGG1 is a DNA glycosylase/apurinic/apyrimidinic (AP) lyase that is involved in base excision repair for eliminating 8-oxo-G from DNA. In this study, the functions of AtOGG1 were elaborated. 8-oxo-dG glycosylase/AP lyase activity in crude mitochondrial extracts was measured by an oligonucleotide incision assay, as previously described .
